Eh... probably not. II is set 500 years after I, so there isn't really much chance that there'll be much to be lost on. You might miss out on little references to Fable I, but nothing that will challenge your enjoyment or understanding of the game.

By all means, play it (I'd suggest TLC), it's cool... but with II coming out tomorrow/three days from now you don't have much time to do it. Might as well hit Fable II, which should be better anyway.
