I have to have sex 25 times. Got nothing wrong with that but I have a question.

Can I invite, let's say, 6 people at once and have sex with them and for it to count as 6 times?

Nah, every time you see the black screen of love that counts as 1 time, BTW you need to make LOVE, not sex, so prostitutes are out of the question. You may only sleep with somone you married to.

Yeah, just have do it once more and you're good. Just no prostitutes.

When I got the download my character had already done the requirements so it just activated automatically

The achievement should unlock immediately after loading the profile that has slept with over 25 people - provided you've downloaded the XBL update too, that is. Angel got the achievement just for loading up the game because she'd already exceeded that number of required conquests. The same goes the Demon Doors achievement, if anyone's curious. She unlocked that, too, upon loading up her character.
